Culture to Transfection-Ready DNA in Eight Steps
1. Harvest: 4°C / 5000 rpm / 15 min; resuspend in endotoxin-free PT.
2. Lyse & Clear RNA: RNase A at 100 µg/mL; Proteinase K optional.
3. Neutralize: Buffer P3/ER, BenzoNuclease for dense cultures.
4. Capture: Load onto a PLC column or self-packed PLR18 column.
5. Wash & Elute: Gravity throughout, run many columns in parallel.
6. Concentrate: Isopropanol precipitation + 70% ethanol wash.
7. Quality Control: A260/A280 1.8–2.0 plus agarose gel.
8. Build (Enzymes): FlashCut digest – SAP01 – LG02 ligate EN01/EN03.
