Description
This product employs colloidal gold lateral flow immunoassay technology to rapidly detect the concentration of His-tagged proteins in cell culture supernatants, cell/bacterial lysates, or purified proteins via a competitive binding mechanism. It constitutes a semi-quantitative His-protein detection card.
The primary structure of this product comprises a sample pad, binding pad, chromatographic membrane, absorbent paper, and backing plate. The chromatographic membrane is coated with goat anti-mouse IgG polyclonal antibody (control line C) and His-tagged protein (test line T). The binding pad is coated with colloidal gold-labelled anti-His-tagged antibody. When the test sample is applied to the sample pad, capillary action draws the liquid towards the binding pad. If the sample does not contain His-tagged protein, the gold-labelled anti-His-tagged antibody on the binding pad binds to the His-tagged protein immobilised at the test line (T), forming an ‘Au-anti-His-tagged antibody-His-tagged protein’ complex. This results in a red band appearing at the test line (T). If the test sample contains His-tagged protein, the His-tagged protein binds to the gold-labelled anti-His-tag antibody on the binding pad, forming an ‘ Au-anti-His-tag antibody-His-tagged protein’ complex. The unreacted gold-labelled anti-His-tag antibody on the binding pad then reacts with the His-tagged protein on the test line T, resulting in a red band appearing on the T line. As the concentration of His-tagged protein in the sample increases, the colour intensity of the T line gradually diminishes until it becomes completely absent. Regardless of the presence of His-tagged protein in the sample, the gold-labelled anti-His-tag antibody on the conjugate pad will migrate upward to the control line C, reacting with the goat anti-mouse IgG to produce a red band. The red band displayed on control line C serves as the criterion for assessing the integrity of the chromatographic process.
