The Super‑Competent Cell Preparation Kit manufactured by our company is designed for rapid preparation of E. coli competent cells with high transformation efficiency. This kit is moderately modified based on conventional super‑competent cell preparation protocols, featuring simple operation and high transformation efficiency.
Using this kit, competent‑cell efficiency can reach 108‑109 cfu/μg plasmid. Higher efficiency is achieved for small plasmids, whereas slightly lower efficiency is observed for large plasmids. Super‑competent cells prepared with this kit are suitable not only for plasmid transformation but also for routine ligation product transformation, especially for blunt‑end ligation applications which demand high transformation efficiency.
The experimental workflow is straightforward. Approximately 60 min are required for competent‑cell preparation after bacterial culture is completed.
This kit is compatible with most common E. coli strains, including DH5α, JM109, TG1, HB101 and XL‑1. It also works for special E. coli strains used for protein expression or viral plasmid construction. Plasmid transformation is reliably achievable with prepared competent cells; nevertheless, transformation performance for ligation products may be inferior compared with strains such as DH5α.
Specially formulated bacterial growth medium dedicated to super‑competent cell preparation is supplied in the kit. It improves transformation efficiency of resultant competent cells and saves users from medium‑reagent preparation.
The kit can be used in multiple batches, providing reagents sufficient for 100 transformation assays in total.
